The important point is: try to be ORIGINAL. Don't copy something you've seen before, especially if it's something popular like "Twilight". In fact, try to avoid vampires altogether (they've been done to death).
Start with a character. What's interesting or special about them? Then, how might this affect their life? This will give you a scene of your story. How do you get the reader there in an interesting way? How does this change things for your character? How do they feel, and what do they do about it?
My ideas just come to me, so I can't really help on the inspiration front. If you're really stuck, pick twenty random words out of a dictionary and try to build a story around them.
Rememer: CHARACTER and ORIGINALITY!
